The Doctor has never twigged to the possibility that River is a Time Lord or rather Time Lady (given how he claims to be able to sense Time Lords, it seems contradictory not to recognise River as one). And yet the girl in the spacesuit shows the ability to regenerate at the end of Day of the Moon.
From what I've heard, the Doctor doesn't twig to the fact that the kid is a Time Lady, but maybe you can't if they haven't regenerated. Fan lore states that Time Lords don't grow a second heart until after their first. Maybe the Doctor can't quite sense another Time Lord until after their first regeneration. And then again, if the girl is an engineered hybrid of Time Lord and human (perhaps explaining why she needs the spacesuit as a life-support system), that could also explain why he couldn't sense her.
I say, River is human. She probably has some relation to Amy, but I doubt that River is Amy's daughter, or a Time Lady.
Whether or not the girl in the spacesuit is Amy's child-in-potentia is another kettle of fish entirely.

I don't mind it. It's certainly better than
The Eleventh Hour as far as season openers go. And as The Mad Collector stated, the Neil Gaiman episode is the fourth one, on in just over three weeks. Supposedly, we will be meeting someone that we haven't seen since
The War Games. That means only three people: Zoe (yes, please!), Jamie, and the War Chief.

Post-hypnotic suggestion from the Silence, I believe that they imply later. The Silence probably said that to Amy (and thus, the Doctor) the Doctor to lure him somewhere, or to impair his ability to fight them.

period. During at least three regenerations, the Doctor actually came close to dying permanently (
Planet of the Spiders, where he needed K'Anpo/Cho-Je's help to regenerate after dying in Sarah's arms;
The Caves of Androzani, where he tells Peri that he might regenerate, but he doesn't know whether he will, as 'it feels different this time'; and the TV Movie, where the Doctor states that he nearly died permanently because of the anaesthetic used in a botched operation to cure what the surgeons thought was a heart condition), and in the novelisation of
Battlefield, while being throttled by a snake made of plasma, the Doctor recalls a Time Lord lecture that states that regeneration is impossible with total brain death.
